The clearance of (+)-[14C]isoproterenol by the liver and the urinary and biliary excretion of radioactivity were determined at different pH values in the range from pH 4.0 to 9.0, using both in situ perfusion and isolated perfused liver techniques. In both cases, the clearance of (+)-[14C]isoproterenol was pH dependent and the overall hepatic clearance of (+)-[14C]isoproterenol decreased as the pH was increased from pH 4.0 to 6.0, followed by an increase to 7.0. In both perfusion and isolated liver studies, the highest hepatic clearance and hence excretion of radioactivity was observed at pH 7.0. The renal clearance of (+)-[14C]isoproterenol was also pH dependent. As with the hepatic disposition, the renal clearance of (+)-[14C]isoproterenol was highest at pH 7.0, although there was only a modest difference between the overall clearance at pH 7.0 and 8.0 in the isolated perfused rat liver studies. The results indicate that the hepatic clearance of (+)-[14C]isoproterenol is influenced by the pH of the perfusate.
